• An adult alpaca (Lama pacos) had a locally extensive area of hepatic atrophy involving the right lobe. Grossly, the atrophic lobe was light tan and firm and contained small, raised, white to yellow, partially mineralized circular nodules predominantly at the periphery of the atrophic tissue. Microscopically, viable hepatocytes were not present in the atrophic area, and the tissue consisted of diffuse biliary epithelial proliferation without any evidence of nuclear or cellular atypia or the presence of mitotic figures. The circular mineralized nodules consisted of granulomatous inflammation with intralesional parasitic ova surrounded by fibrous connective tissue. Morphologically, the ova were compatible with those of Fasciola hepatica. The severe biliary hyperplasia was unusual, and it was not clear whether it was caused by an aberrant host response to the parasitic infection or whether it was an unrelated event. more »
  • Objective—To develop a PCR assay for Candidatus Mycoplasma haemolamae (CMhl) infection in alpacas and use it to study the efficacy of oxytetracycline treatment and development of a subclinical carrier state. Animals—8 healthy adult alpacas. Procedures—Alpacas initially had negative results for CMhl in blood samples via PCR assay and were experimentally infected with CMhl; 4 were treated with oxytetracycline, and 4 were not treated. All were monitored regularly via PCR assay, blood smear examination, PCV, rectal temperature, and physical examination. At 6 months after treatment, all alpacas were immunosuppressed by administration of dexamethasone and tested for CMhl. Results—7 of 8 alpacas had positive PCR assay results 4 to 6 days after experimental infection. When organisms were detectable on a blood smear, they were seen 2 to 6 days after positive results of PCR assay. Infection was often associated with mild anemia that was usually transient. No alpacas became hypoglycemic. Oxytetracycline treatment was not associated with faster clearance of organisms or resolution of anemia, and 4 of 4 treated alpacas still had positive results of PCR assay when immunosuppressed 6 months later; 0 of 3 nontreated alpacas had positive results of PCR assay following immunosuppression. Transient fever was detected in 3 alpacas during immunosuppression. Conclusions and Clinical Relevance—The PCR assay was more sensitive than blood smear examination for detection of infection. Clinical signs, anemia, and fever were not necessarily associated with infection. Oxytetracyline administration did not consistently clear CMhl infection. Although treated with oxytetracycline, infected alpacas remained chronic carriers. more »
  • Three classes of IgG have been described for camelids. IgG1 has a conventional four-chain structure, while IgG2 and IgG3 do not incorporate light chains. The structures and antigen-binding affinities of the so-called heavy-chain classes have been studied in detail; however, their regulation and effector functions are largely undefined. The aim of this study was to examine the participation of conventional and heavy-chain IgG antibodies in the camelid immune defense directed against West Nile virus (WNV). We found that natural infection or vaccination with killed WNV induced IgG1 and IgG3. Vaccination also induced IgG1 and IgG3; IgG2 was produced during the anamnestic response to vaccination. When purified IgGs were tested in plaque-reduction neutralization titer (PRNT) tests, IgG3 demonstrated PRNT activities comparable to those of conventional IgG1. In contrast, IgG2 demonstrated only suboptimal activity at the highest concentrations tested. Flow cytometric analysis revealed that macrophages bound IgG1, IgG2, and IgG3. Furthermore, subneutralizing concentrations of all three isotypes enhanced WNV infection of cultured macrophages. Our results document distinctions in regulation and function between camelid heavy-chain isotypes. The reduced size and distinct structure of IgG3 did not negatively impact its capacity to neutralize virus. In contrast, IgG2 appeared to be less efficient in neutralization. This information advances our understanding of these unusual antibodies in ways that can be applied in the development of effective vaccines for camelids. more »
  • Melanin plays important roles in the formation of animal hair color, the members of TYR gene family participate in the synthesis of melanin. For exploring the relationship between gene expression of alpaca TYR gene family and alpaca's coat color, the relative expression quantity of TYR,TRP1,TRP2 in alpaca of different colors were analyzed by using real-time quantitative PCR in this research. Results showed that the relative expression quantity of TYR,TRP1,TRP2 in Brown alpaca respectively were 13.669,3.417,8.593 times than that in White alpaca, all results were corrected by the household gene. The findings indicated that gene expression level of TYR gene family in Brown alpaca were higher than that in White alpaca, and the gene expression level of TYR gene family were related with phenotype of alpacas' coat color. more »
  • Pacomarca is an experimental ranch founded by the INCA group to act as a selection nucleus from which basic genetic improvement of alpaca fibre can spread throughout the rural communities in the Peruvian Altiplano. State-of-art techniques in animal science, such as performance recording or assisted reproduction including embryo transfer, are applied to demonstrate their usefulness in the Altiplano conditions. Pacomarca has developed useful software (Paco Pro) to carry out the integral processing of production and reproduction data. Mating is carried out individually, and gestation is diagnosed via ultrasound. Breeding values estimated from a modern genetic evaluation are used for selection, and embryo transfer is applied to increase the selection intensity. However, the objective of Pacomarca goes beyond, extending its advances to the small rural communities. Training courses for farmers are organised while searching for new ways of improving the performance of alpacas both technically and scientifically. more »
